چکیده

L-Fucose (6-deoxy-L-galactose) is widely distributed in nature as a component of serum glycoproteins (a), the blood group substances (3), and many plant (4) and bacterial polysaccharides (5). The pathway of biosynthesis of this compound is unknown. Dudman and Wilkinson (5) reported that Aerobacter cloacae NCTC 5920 produces a polysaccharide which contains approximately equal quantities of L-fucose, D-glucose, D-galactose, and uranic acid. In the present studies, this organism was grown on specifically labeled glucose as the sole source of carbon. The distribution of isotope in the L-fucose was determined and compared with the distribution in n-glucose and Dgalactose of the polysaccharide. The results of these experiments indicate that the specifically labeled n-glucose carbon chain is converted to that of L-fucose, n-glucose, or Dgalactose without significant inversion or randomization of the isotope.

Control of colanic acid synthesis.

The nucleotide pools of certain mucoid, colanic acid-synthesizing strains of Escherichia coli, Salmonella typhimurium, and Aerobacter cloacae were examined, and in all cases the nucleotide sugars uridine-5'-diphosphate glucose (UDPG), uridine-5'-diphosphate galactose (UDPGal), guanosine-5'-diphosphate fucose (GDPF), and uridine-5'-diphosphate glucuronic acid (UDPGA) were detected. It is postula...

Formation of guanosine diphosphate L-fucose from guanosine diphosphate D-mannose.

The 6-deoxyhexose, L-fucose, has a wide distribution in nature as a component of bacterial, plant, and animal polysaccharides or oligosaccharides. Its configuration, which is the same as Lgalactose, suggested to earlier workers that it might be formed biosynthetically from n-galactose by reduction of C-l to a methyl group and oxidation of C-6 to an aldehyde. This suggestion was rendered untenab...

The biosynthesis of cell wall lipopolysaccharide in Escherichia coli. V. Purification and properties of 3-deoxy-D-manno-octulosonate aldolase.

3-Deoxy-D-manno-octulosonate aldolase, an inducible enzyme isolated from extracts of 3-deoxy-D-manno-octulosonate-grown Aerobacter cloacae, has been purified approximately 60-fold. The enzyme catalyzes the following reaction: 3-deoxy-D-manno-octulosonate -2 pyruvate + D-arabinose. Pyruvate was characterized chromatographically and with lactic acid dehydrogenase. D-Arabinose was characterized ch...

Structural studies on colanic acid, the common exopolysaccharide found in the enterobacteriaceae, by partial acid hydrolysis. Oligosaccharides from colanic acid.

The exopolysaccharide slime colanic acid has been isolated from representative strains of Escherichia coli, Salmonella typhimurium and Aerobacter cloacae. Analysis showed that each polymer contained glucose, galactose, fucose and glucuronic acid, together with acetate and pyruvate. The molar proportions of these components were 1:1.8:1.9:1:1:1 approximately. On the basis of periodate oxidation ...

Purification and properties of 2-ketogluconokinase from Aerobacter aerogenes.

A specific kinase for the phosphorylation of 2-ketogluconate with adenosine triphosphate has been found in Pseudomonas JEuorescens grown on glucose (l), in Aerobacter cloacae grown on 2-ketogluconate (2), and in a number of other bacterial species. In P. jluorescens, phosphorylation of 2-ketogluconate is a major step in the sequence of glucose catabolism (3) and hence is induced by growth on gl...
